Glenister was born in Harrow, Middlesex, and raised in Hatch End. He is the son of director John Glenister and Joan Fry Lewis, and has a brother, actor Robert Glenister. He attended Hatch End High School. In his early career, Glenister worked as a runner for the Robert Stigwood Organisation, later transitioning to a role as a film publicist until he was 23. His interest in acting was piqued after seeing his brother perform in a student play about the Sex Pistols. Encouraged by his sister-in-law, Amanda Redman, he decided to pursue acting and enrolled at the Central School of Speech and Drama.
