Hamilton played in 13 games as a freshman, earning one start, and finished with 41 tackles, four interceptions, and a touchdown. His performance earned him Freshman All-American honors from the Football Writers Association of America.

As a sophomore in 2020, Hamilton took on a starting role. His first interception of the season came in the 2020 ACC Championship Game against Clemson. He was named First-Team All-ACC after registering 63 tackles, six pass breakups, and one interception. In 2021, Hamilton continued as a starter, but a knee injury sidelined him after just seven games.
